Glover iniziò la sua carriera a Broadway verso la fine degli anni ’60, cosa che gli permise di avere delle piccole parti in alcuni film degli anni ’70. Negli anni ’80 interpretò il ruolo del cattivo in molti film, tra cui “The Evil That Men Do” e  “52 Pick-Up”. John è stato anche nominato algi Emmy un paio di volte, grazie alle sue interpretazioni nelle serie “An Early Frost” (1985) e  “L.A. Law”. Ha preso inoltre parte a molti telefilm (come Miami Vice, La signora in giallo, Frasier e Smallville) e a film quali “Gremlins 2” e “Robocop 2”.

Abile uomo d'affari dal passato quantomeno discutibile, è stato processato con l'accusa di aver commissionato l'omicidio dei suoi genitori. Vedovo, sua moglie, morta per leucemia, ha ucciso il loro stesso secondogenito perchè non dovesse avere un padre come lui. Padre incapace di affetto e crudele, ogni suo tentativo di instaurare un rapporto con il suo primogenito, Lex, si è sempre rivelato altrimenti; ne ha addirittura commissionato l'omicidio, assieme a quello di Chloe Sullivan. Fa condurre discutibili esperimenti genetici all'interno di livelli segreti nelle sue fabbriche. costantemente alla ricerca di un modo per sottomettere suo figlio, e chiunque incontro il suo cammino, si è spinto fino alla clonazione umana. Ispirerebbe sfiducia ad un serpente a sonagli, la cosa che gli è più congeniale è il ricatto, per questo si è guadagnato un astio nei suoi confronti che però non gli rende pienamente giustizia.
